Premium business class passengers will be able to book transport to or from the airport to surrounding communities in the city free of charge.
In order to take advantage of the new deal, passengers are required to call a special Czech Airlines customer service line.
Petre Pistelak, the airline’s vice president for marketing and development, said: "Part of Czech Airlines’ long-term business strategy has been to focus on business travellers – those who fly regularly and expect a high level of service.
"This complementary airport shuttle service is a new addition to the already extensive range of benefits that Czech Airlines offers its Business Premium class travellers."
However, the service is restricted to those passengers with business premium tickets on OK coded flights.
Authorities at Prague Airport recently announced their intention to invest more than CZK2.3 billion (74.6 million) in the Ruzyne Airport ahead of its planned privatisation next year.
