Kevin Healy, the airline’s senior vice president of marketing and planning, said the new flights would connect West Virginia’s capital city with "one of the family vacation capitals of the world".
"Our expansion to Charleston opens a gateway for residents in the heart of the Appalachians to get to Orlando, home of America’s favourite vacation destination," he commented.
Yeager Airport director Rick Atkinson said he was "very excited" to welcome AirTran Airways, which he referred to as "a true low-cost carrier".
The addition of the Charleston route means that the airline now offers flights to 63 destinations throughout the US, including 36 non-stop destinations from Orlando.
AirTran Airways was recently named the top low-cost airline by the Airline Quality Rating.
The accolade was achieved for the second year in succession, with Honolulu-based Hawaiian Airlines coming first overall.
